Functionalized Surfaces as a Tool for Virus Sensing: A Demonstration of Human mastadenovirus Detection in Environmental Waters

The main goal of this study was to apply magnetic bead surface functionalization in the form immunomagnetic separation (IMS) combined with real-time polymerase chain reaction (qPCR) (IMS-qPCR) detect Human mastadenovirus species C (HAdV-C) and F (HAdV-F) water samples. technique efficiency compared a nonfunctionalized method (ultracentrifugation) followed by laboratory detection. Tests were carried out standardize IMS parameters tests on 15 samples concentrated ultracentrifugation. Microscopic analyses detected successful beads–antibody attachment. HAdV up dilutions 10−6 IMS-qPCR, able infect cell cultures. In samples, HAdV-C 60% (monoclonal) 47% (polyclonal) while 13% ultracentrifugation gave positive result. HAdV-F 27% IMS-qPCR 20% (monoclonal). rate detection varied from 4.55 × 102 5.83 106 genomic copies/L for 2.00 2.11 103 GC/L showed be more effective concentration than ultracentrifugation, improving assessment infectious resources.
